Source

Human TPK1, His Tag (TP1-H5146) is expressed from E.coli cells. It contains AA Glu 2 - Ser 243 (Accession # AAH68460).

Predicted N-terminus: Met

Molecular Characterization

Poly-his
TPK1(Glu 2 - Ser 243)AAH68460

This protein carries a polyhistidine tag at the N-terminus.

The protein has a calculated MW of 28 kDa. The protein migrates as 28 kDa under reducing (R) condition (SDS-PAGE).

Endotoxin

Less than 1.0 EU per μg by the LAL method.

Purity

>95% as determined by SDS-PAGE.

Formulation

Lyophilized from 0.22 μm filtered solution in 50 mM Tris, 150 mM NaCl, pH8.0. Normally trehalose is added as protectant before lyophilization.

Reconstitution

Please see Certificate of Analysis for specific instructions.

For best performance, we strongly recommend you to follow the reconstitution protocol provided in the CoA.

Storage

For long term storage, the product should be stored at lyophilized state at -20°C or lower.

Please avoid repeated freeze-thaw cycles.

No activity loss is observed after storage at:

  1. 4-8°C for 12 months in lyophilized state;
  2. -70°C for 3 months under sterile conditions after reconstitution.

SDS-PAGE

Human TPK1, His Tag (Cat. No. TP1-H5146) SDS-PAGE gel

Human TPK1, His Tag on SDS-PAGE under reducing (R) condition. The gel was stained overnight with Coomassie Blue. The purity of the protein is greater than 95%.

Background

Thiamin pyrophosphokinase 1 (TPK1),a member of the thiamine pyrophosphokinase family, is also known as Thiamine pyrophosphokinase 1, Placental protein 20 (PP20) and hTPK1,which is detected in heart, kidney, testis, small intestine and peripheral blood leukocytes, and at very low levels in a variety of tissues.TPK1 is regarded as a Kinase and a Transferase that can bind ATP and Nucleotide. TPK1 catalyzes the phosphorylation of thiamine to thiamine pyrophosphate and can also catalyze the phosphorylation of pyrithiamine to pyrithiamine pyrophosphate. As for disease,TPK1 is relevant to thiamine metabolism dysfunction syndrome 5, episodic encephalopathy type (THMD5).
