Source

Recombinant Human CLIC4 Protein (rh CLIC4) Met 1 - Lys 253 (Accession # AAH12444) was produced in E.coli cells at ACROBiosystems.

Molecular Characterization

rh CLIC4 is fused with a polyhistidine tag at the N-terminus, and has a calculated MW of 29.6 kDa. The predicted N-terminus is Met. DTT-reduced Protein migrates as 25-30 kDa in SDS-PAGE .

Endotoxin

Less than 1.0 EU per μg of the rh CLIC4 by the LAL method.

Purity

>92% as determined by SDS-PAGE.

Formulation

Lyophilized from 0.22 μm filtered solution in PBS, pH 7.4. Normally Mannitol or Trehalose are added as protectants before lyophilization.

Reconstitution

See Certificate of Analysis for reconstitution instructions and specific concentrations.

Storage

Avoid repeated freeze-thaw cycles.

No activity loss was observed after storage at:
In lyophilized state for 1 year (4oC); After reconstitution under sterile conditions for 3 months (-70oC).

SDS-PAGE

Recombinant Human CLIC4 Protein
The purity of rh CLIC4 was determined by DTT-reduced (+) SDS-PAGE and staining overnight with Coomassie Blue.

Background

Chloride intracellular channel protein 4 (CLIC4) ,a member of the chloride channel CLIC family, is also known as intracellular chloride ion channel protein p64H1,which contains one GST C-terminal domain. CLIC4 can insert into membranes and form poorly selective ion channels that may also transport chloride ions. Channel activity depends on the pH. Membrane insertion seems to be redox-regulated and may occur only under oxydizing conditions. Also,CLIC4 can promote cell-surface expression of HRH3, endothelial cell proliferation and regulate endothelial morphogenesis. Furthermore ,CLIC4 has alternate cellular functions like a potential role in angiogenesis or in maintaining apical-basolateral membrane polarity during mitosis and cytokinesis.
