Human Kininogen 1 / KNG1 Protein

  • Synonym
    KNG1,Kininogen-1,HMWK,BDK,KNG
  • Source
Human Kininogen 1, His Tag (KN1-H5225) is expressed from human 293 cells (HEK293). It contains AA Gln 19 - Ser 427 (Accession # NP_000884).
    Predicted N-terminus: Gln 19
  • Molecular Characterization
    Kininogen 1(Gln 19 - Ser 427) NP_000884

This protein carries a polyhistidine tag at the C-terminus.

The protein has a calculated MW of 46.6 kDa. The protein migrates as 50-68 kDa under reducing (R) condition (SDS-PAGE) due to glycosylation.

  • Endotoxin
Less than 1.0 EU per μg by the LAL method.
  • Purity

    >92% as determined by SDS-PAGE.

  • Formulation

Lyophilized from 0.22 μm filtered solution in 100 mM Sodium Acetate, 100 mM NaCl, pH5.5. Normally trehalose is added as protectant before lyophilization.

Contact us for customized product form or formulation.

  • Reconstitution

Please see Certificate of Analysis for specific instructions.

For best performance, we strongly recommend you to follow the reconstitution protocol provided in the CoA.

  • Storage

For long term storage, the product should be stored at lyophilized state at -20°C or lower.

    Please avoid repeated freeze-thaw cycles.

No activity loss is observed after storage at:

    1. 4-8°C for 12 months in lyophilized state;
    2. -70°C for 3 months under sterile conditions after reconstitution.
SDS-PAGE
Human Kininogen 1, His Tag (Cat. No. KN1-H5225) SDS-PAGE gel

Human Kininogen 1, His Tag on SDS-PAGE under reducing (R) condition. The gel was stained overnight with Coomassie Blue. The purity of the protein is greater than 92%.

  • Background
Kininogen-1 (KNG1) is also known as HMWK, BDK, KNG, is a secreted protein which contains three cystatin domains. KNG1 can be cleaved into the following six chains: Kininogen-1 heavy chain, T-kinin, Bradykinin, Lysyl-bradykinin, Kininogen-1 light chain, Low molecular weight growth-promoting factor. HMW-kininogen plays an important role in blood coagulation by helping to position optimally prekallikrein and factor XI next to factor XII; HMW-kininogen inhibits the thrombin- and plasmin-induced aggregation of thrombocytes. LMW-kininogen inhibits the aggregation of thrombocytes; LMW-kininogen is in contrast to HMW-kininogen not involved in blood clotting.
