  • Background
Human Interleukin-8 (IL-8) OneStep ELISA Kit is a one-step quick immunoassay designed to measure human IL-8 levels in cell culture supernates, serum, and plasma. This assay kit is a standard sandwich-ELISA format. It contains recombinant human IL-8 and a pair of antibodies against the recombinant factor, which are provided by ACROBiosystems. Results are obtained by four parameter logistic curve that were parallel to the standard curves obtained. The verification results indicate that this kit can be used for the quantitative determination of natural and recombinant human IL-8 concentrations. The specificity has been verified.
  • Application

The kit is developed for quantitative detection of IL-8 in human serum and cell culture supernates.

It is for research use only.

  • Reconstitution
Please see Certificate of Analysis for details of reconstitution instruction and specific concentration.
  • Storage
The unopened kit is stable for 12 months from the date of manufacture if stored at 2°C to 8°C.

The opened kit should be stored per components table. The shelf life is 30 days from the date of opening.

  • Assay Principles
This assay kit is used to measure the concentrations of human Interleukin 8 (IL-8) by employing a standard sandwich-ELISA format. The microplate in the kit has been pre-coated with Anti- IL-8 Antibody. First add the standard samples provided in kit and your samples to the plate, next add detection antibody Biotin-Anti-IL-8 Antibody to the plate, incubate and wash the wells. After wash add HRP-Streptavidin to the plate, incubate and wash the wells. Lastly load the substrate into the wells and color develops in proportion to the amount of Interferon-γ bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450nm and 630nm.
Typical Data Please refer to DS document for The assay protocol.
 IL-8 ELISA

For each experiment, each ELISA plate needs to set the standard curve. The minimum detectable concentration of CRS-B004 is less than 5.0 pg/mL.

  • Background: IL-8
    Interleukin 8, IL-8
IL-8 is a chemotactic factor that attracts neutrophils, basophils, and T-cells during the inflammatory process. Like all chemokines, IL-8 is a small, soluble peptide (8–10 kDa).Interleukin-8 is synthesized primarily as a 99 amino acid peptide that is processed through signal sequence cleavage and N-terminal proteolysis to yield several active IL-8 isoforms , with peptides of 72 and 77 amino acids being most biologically relevant. The 72 amino acid peptide is the major form secreted by monocytes and macrophages in culture, whereas the 77 amino acid variant is the most abundant secretory product of nonimmune cells.
