  • Product Overview
    T helper cells (Th) play a vital role in the adaptive immune system by suppressing or regulating immune responses. After Th actived, it will differentiate into T-helper 1 (Th1) or T-helper 2 (Th2) effector cells. These two subsets are characterized by their distinct cytokine expression pattern and the immune function they mediate. Th1 and Th2 cells each produce a specialized combination of cytokines that depends on type of pathogen that the helper T cell has recognized—some cytokines are more effective than others in the process of eliminating certain invaders. In general, TH1 cytokines play the role of immune killing, while Th2 cytokines play the role of immune protection. Th1/Th2 paradigm provides rationale for developing new vaccines against infectious diseases and novel strategies for the therapy of allergic and autoimmune disorders.

    Mouse Th1/Th2 Cytokine Kit (6-plex) (Flow Cytometry Multiplex Bead Assay, Non-Magbeads) gives quantitative results of f Interleukin-2 (IL-2), Interleukin-4 (IL-4), Interleukin-6 (IL-6), Interleukin-10 (IL10), Tumor Necrosis Factor α type (TNF-α) and Interferon-γ (IFN-γ) with a sample testing. The performance of this kit has been optimized for specific analysis of cytokines in mouse serum samples.

    It is for research use only.

  • Storage
    The unopened kit is stable for 12 months from the date of manufacture if stored at 2°C to 8°C.

    The opened kit should be stored per components table. The shelf life is 30 days from the date of opening.

  • Assay Principles
    Mouse Th1/Th2 Cytokine 6-plex panel is a multiplex bead-based indirect immunofluorescence assay, utilizing flow cytometry for data acquisition and classification. The beads conjugated with specific antibody, target protein (mouse IL-2/4/6/10, TNF-α, IFN-γ) in specimens and Biotin-conjugated detection antibody form a sandwiches complex as bead-analyte-detection antibody. Streptavidin-phycoerythrin (SA-PE) is subsequently added and would bind to biotinylated detection antibodies. The intensity of PE fluorescence signal in proportion to the target protein in specimens, are assessed by flow cytometry. The present assay has been optimized for quntitatively analysis for the mouse IL-2/4/6/10, TNF-α, IFN-γ in mouse serum samples.

 Th1/Th2 cytokine TYPICAL DATA

A double logarithmic fitting curve was plotted for each cytokine ( IL-2,IL-4,IL-6, IL-10, TNF-α, IFN-γ). The present curve of each cytokines ( IL-2,IL-6, IL-10, TNF-α, IFN-γ) was ranging from 9.8 pg/mL to 10000 pg/mL respectively; The present curve of IL-4 was ranging from 9.8 pg/mL to 5000 pg/mL.
