Biotinylated SARS-CoV-2 Spike RBD Protein (L452R, T478K), His,Avitag (SPD-C82Ed) is expressed from human 293 cells (HEK293). It contains AA Arg 319 - Lys 537 (Accession # QHD43416.1 (L452R, T478K)). The mutations L452R, T478K were identified in the SARS-CoV-2 Delta variant (Pango lineage: B.1.617.2; other names: 21A/S:478K).
Predicted N-terminus: Arg 319

This protein carries a polyhistidine tag at the C-terminus, followed by an Avi tag (Avitag™).
The protein has a calculated MW of 28.3 kDa. The protein migrates as 33-40 kDa under reducing (R) condition (SDS-PAGE) due to glycosylation.
Labeling
Biotinylation of this product is performed using Avitag™ technology. Briefly, the single lysine residue in the Avitag is enzymatically labeled with biotin.
Protein Ratio
Passed as determined by the HABA assay / binding ELISA.
Purity
>90% as determined by SDS-PAGE.
Formulation
Lyophilized from 0.22 μm filtered solution in PBS, pH7.4 with trehalose as protectant.

We placed an order for Omicron RBD, to run side-by-side analyses of our neutralizing antibodies against Omicron vs Wild type/Wuhan RBD protein (also from Acro) in biolayer interferometry experiments. AcroBio's Biotinylated Omicron RBD protein loads onto streptavidin biosensors very well - achieving a 1nm spectral shift within 40 seconds using a 100nM solution. We were able to obtain satisfactory results that were consistent with findings from other orthogonal methods for characterizing our antibodies (ELISA, viral infectivity, etc). Protein arrived in a timely manner.
