  • Background
    Adeno-associated virus (AAV) is the simplest and non-enveloped single-stranded DNA virus with a viral genome length of about 4.7Kb, which belongs to the parvovirus family. AAV has become one of the most important gene vectors in the field of gene therapy due to its long-term expression, low toxicity, low immunogenicity, and high tissue specificity.
  • Application

    AAV Titration Detection Kit (Universal )was developed for the detection and quantitative determination of AAV capsid titration in AAV gene therapy product preparation processing.

    It is for research use only.

  • Reconstitution
    Please see Certificate of Analysis for details of reconstitution instruction and specific concentration.
  • Storage
    The unopened kit is stable for 36 months from the date of manufacture if stored at -20°C to -70°C.

    The opened kit should be stored per components table. The shelf life is 90 days from the date of opening.

  • Assay Principles
    This assay employs a standard sandwich-ELISA format, providing a rapid detection of AAV Capsid. The pair consists of AAV Capsid Capture Antibody and AAV CapsidDetection Antibody and HRP-Conjugated Antibody.

    Your experiment will include 5 simple steps:

    a) Coat the plate with AAV Capsid Capture Antibody.

    b) Add your molecule of interest to the tests.

    c) Add AAV Capsid Detection Antibody.

    d) Add HRP-Conjugated Antibody followed by TMB or other colorimetric HRP substrate.

    e) Stop the substrate reaction by add diluted acid. Absorbance (OD) is calculate as the absorbance at 450 nm minus the absorbance at 630 nm to remove background prior to statistical analysis.

 AAV TYPICAL DATA

Immobilized AAV Capsid Capture Antibody (AAVX01-C01) at 1.0 μg/mL (100 μL/well) can bind AAV2 Capsid, and then add AAV Capsid Detection Antibody (AAVX01-C02) at 0.5 μg/mL (100 μL/well), final detection was performed using HRP-Conjugated Antibody (AAVX01-C03) with a linear range of 3.91E+07 - 2.50E+09 capsids/mL (QC tested).

 AAV TYPICAL DATA

Immobilized AAV Capsid Capture Antibody (AAVX01-C01) at 1.0 μg/mL (100 μL/well) can bind AAV Capsid, and then add AAV Capsid Detection Antibody (AAVX01-C02) at 0.5 μg/mL (100 μL/well), final detection was performed using HRP-Conjugated Antibody (AAVX01-C03).

  • Background: AAV
    Through alpaca immunity and phage display, a strain of nanobodies was screened. It binds specifically to native and recombinant adeno-associated virus (AAV) particles of various serotypes with high affinity, including AAV1/2/3/5/6/7/8/DJ. By fusing it with mouse Fc fragments, the antibody pairs that can specifically bind to AAV particles of different serotypes were obtained for the quantitative detection of AAV capsid titers.
