Source

Biotinylated Monoclonal Anti-G4S linker Antibody, Rabbit IgG (016), CF (carrier free), premium grade is a rabbit monoclonal antibody (labelled by chemically labeling technology) recombinantly expressed from human 293 cells (HEK293), it shows superior performance in various applications such as ELISA assays and flow cytometry assays with higher affinity and better specificity.

It is produced under our rigorous quality control system that incorporates a comprehensive set of tests including sterility and endotoxin tests. Product performance is carefully validated and tested for compatibility for cell culture use or any other applications in the early preclinical stage.
GMP-MG4S25 is the GMP version of this G4S-BLM25A1. These two proteins display indistinguishable performance profiles, thereby ensuring a seamless transition for end users from early preclinical stage to later clinical phases.

Application

1. Flow Cytometry (Evaluation of cell surface expressed CARs of varying specificity containing a G4S linker within the scFv of the extracellular domain).

2. Immunoassay of bispecific antibody.

3. CAR T cell enrichment and sorting.

Specificity

It can specifically recognize proteins or ScFv containing (G3S)n or (G4S)n linkers, where n = 2, 3, 4... That is, it can recognize linkers with a minimum length of 2 repeats for both (G3S)n and (G4S)n.

Quality Description

ACRO's Research-grade products are suitable for a wide range of cell culture applications, particularly for research use in academic institutions. These products are sterilized by filtration, followed by lyophilization where applicable. Typical specifications include endotoxin levels of <0.01 EU/μg and purities >95%. Biological activity is calibrated against WHO/NIBSC standards when available.
ACRO's Premium-grade (Pre-GMP) products are characterized by their high quality and enhanced safety profiles, making them ideal for early-stage discovery and manufacturing processes in cell therapy companies. A key advantage is their seamless transition to corresponding GMP-grade versions. Biological activity is calibrated against WHO/NIBSC standards when available. Typical specifications include endotoxin levels of <0.01 EU/μg and purities >95%. In addition, rigorous testing is conducted to ensure the absence of mycoplasma, HCD, and HCP, thereby guaranteeing product safety.

Background

The poly-Glycine-Serine (G4S) linker is a type of flexible, unstructured synthetic peptide linker sequence often leveraged to connect the variable heavy (VH) domain and variable light (VL) domain of single-chain variable fragments (scFvs) and chimeric antigen receptors (CARs) that utilize an extracellular domain scFv for target antigen recognition. The linker itself consists of a core pentapeptide sequence, Gly-Gly-Gly-Gly-Ser, that is repeated and commonly found as either a 15-mer (G4S)3 or20-mer(G4S) 4 within scFv-based CARs and scFv fragments. The linker sequence length plays a role in controlling scFv stability and the noncovalent association between the VH and VL domains.
