Background

HER2 encodes Erb-B2 receptor tyrosine kinase 2, a receptor tyrosine kinase that acts as a coreceptor in EGF receptor family heterodimers and drives cell growth and survival. It has no ligand-binding domain of its own, but it binds tightly to other ligand-bound family members to stabilize ligand engagement and amplify downstream signaling. This signaling role places HER2 at the center of pathways linked to MAPK and PI3K activity.
