Product Description
Anti-G4S Linker NanoSort MicroBeads, Research Grade are dextran-coated superparamagnetic iron oxide nanoparticles conjugated with recombinant monoclonal antibodies specific for the G4S linker (isotype: rabbit IgG). They are designed for the positive selection and enrichment of G4S-containing CAR-expressing cells.
Anti-G4S Linker NanoSort MicroBeads, Research Grade are produced under sterile manufacturing conditions (ISO 5), and no animal- or human-derived components are used throughout the production process. It is produced under our rigorous quality control system that includes a comprehensive set of tests including sterility and endotoxin tests.Application
Cellectra™ Anti-G4S Linker NanoSort MicroBeads, Research Grade are designed for the positive selection and enrichment of CAR-modified cells expressing a G4S linker during CAR-T cell manufacturing following gene transduction. Anti-G4S Linker specifically recognizes the G4S (GGGGS) linker, a flexible peptide widely used in CARs and other recombinant proteins. Unlike antigen-specific reagents, it targets the conserved G4S sequence, providing a universal, antigen-independent solution for the detection, enrichment, and isolation of G4S-containing engineered cells. Its compatibility with diverse CAR constructs simplifies reagent development, streamlines workflows, and ensures consistent performance in research and cell manufacturing applications. The MicroBeads are conjugated with monoclonal antibodies specific for the G4S linker, enabling selective labeling and rapid magnetic enrichment of G4S-containing CAR-expressing cells using magnetic separation columns.
Storage
For long term storage, the product should be stored at liquid state at -20°C or below. Stability upon receipt: • Stable for 24 months at -20 °C or below; • Stable for 8 months at 2–8 °C under sterile conditions.

Consistent enrichment of Anti-CD22 CAR-T cells across a broad range of initial CAR-positive frequencies using Cellectra™ Anti-G4S Linker NanoSort Microbeads.
Separation performance was evaluated using 1 × 10⁷ Anti-CD22 CAR-T cell samples containing initial CAR-positive frequencies ranging from 5% to 60%. Cell separation was performed using 20 μL of Cellectra™ Anti-G4S linker NanoSort Microbeads, Research grade (Cat. No. MBS-S009). Positive and negative fractions were stained with FITC-Labeled Human Siglec-2 (CD22) Protein, Fc Tag and 7-AAD, followed by flow cytometric analysis. Across all tested input frequencies, the isolated fraction consistently achieved high CAR-positive purity (94.76%–98.80%), while residual CAR-positive cells in the negative fraction remained low (0.02%–1.77%). These results demonstrate excellent separation performance, enabling consistently high-purity enrichment of Anti-CD22 CAR-T cells from samples containing 5%–60% CAR-positive cells while maintaining minimal target-cell carryover in the negative fraction.

Stability of Cellectra™ Anti-G4S Linker NanoSort Microbeads Following Freeze–Thaw Cycling and Accelerated Aging at 37℃
Freeze-thaw stability was verified by subjecting the microbeads to 3 cycles prior to separation testing. Comparative analysis against the standard batch confirmed that purity and cell recovery were maintained at baseline specifications, demonstrating robust stability. To ensure prolonged shelf life, storage at -20°C or below is recommended upon receipt (however, repeated freeze-thaw cycles are not advised for routine operational use).
Accelerated aging studies were conducted at 37°C to evaluate long-term stability profiles. Following 3.5 and 7-day incubation periods, the microbeads sustained high purity and cell recovery performance during Anti-CD22 CAR-T cells separation. These data substantiate product stability and validate the established shelf-life specifications.
