Description
The Human BDCA2 (Luc) Jurkat Reporter Cell was engineered to not only express NFAT signaling response element, but also express the receptor full length human BDCA2 (Uniprot: Q8WTT0-1). When cocultured with anti-human BDCA2 agonist antibody, the interaction of agonist antibody and BDCA2 on the surface of Human BDCA2 (Luc) Jurkat Reporter Cell results in NFAT-mediated luminescence.

Application

Agonistic activity analysis of anti-human BDCA2 antibody (RLU).
This reporter cell was incubated with serial dilutions of antibodies in the presence of CHO cells or CHO/Human CD32b Stable Cell Line (Medium Expression) (Cat. No. SCCHO-ATP060M). The EC50 of Litifilimab determined in the presence of CHO cells was approximately 0.4915 μg/mL, and in the presence of CHO/Human CD32b Stable Cell Line (Medium Expression) was approximately 0.1866 μg/mL. These results demonstrate that Litifilimab can activate BDCA2 signaling independent of CD32b-mediated crosslinking, but the presence of CD32b can enhance BDCA2 signaling.Protocol

This reporter cell was incubated with serial dilutions of antibodies in the presence of CHO cells or CHO/Human CD32b Stable Cell Line (Medium Expression) (Cat. No. SCCHO-ATP060M). The max induction fold of Litifilimab determined in the presence of CHO cells was approximately 81.03, and in the presence of CHO/Human CD32b Stable Cell Line (Medium Expression) was approximately 190.72. These results demonstrate that Litifilimab can activate BDCA2 signaling independent of CD32b-mediated crosslinking, but the presence of CD32b can enhance BDCA2 signaling.Protocol
