Source

Biotinylated Human LC3B Protein, Avitag,His Tag (LCB-H82Q3) is expressed from E. coli cells. It contains AA Met 1 - Val 125 (Accession # Q9GZQ8).

Predicted N-terminus: Met

Molecular Characterization

This protein carries an Avi tag (Avitag™) at the N-terminus, followed by a polyhistidine tag.

The protein has a calculated MW of 18.4 kDa. The protein migrates as 19-20 kDa when calibrated against Star Ribbon Pre-stained Protein Marker under reducing (R) condition (SDS-PAGE).

Labeling

Biotinylation of this product is performed using Avitag™ technology. Briefly, the single lysine residue in the Avitag is enzymatically labeled with biotin.

Protein Ratio

0.7-1.0 as determined by the SABA assay / HABA assay / binding ELISA.

Purity

>90% as determined by SDS-PAGE.

>95% as determined by SEC-MALS.

Formulation

Lyophilized from 0.22 μm filtered solution in 20 mM Citric acid, 200 mM NaCl, pH3.5 with trehalose as protectant.

Background

LC3B is a pivotal protein encoded by the MAP1LC3B gene, widely recognized as a core component and indicator of cellular autophagy. It plays an essential role in autophagosome formation and the regulation of the autophagic degradation pathway. Critically, the conversion of cytosolic LC3B-I to lipidated, autophagosome-associated LC3B-II upon autophagy activation serves as a robust and widely used biochemical hallmark for assessing autophagic activity.
