Solutions for T Cell Activation and Culture
Cell therapy is the next big step for immuno-oncology, and T cells play a big role in bringing this therapy to patients across the role. Finding the right reagents to isolate, develop, or manufacture T cells while ensuring them to be compliant with the relevant regulations can be a challenge. Currently, the most common types of cell therapies utilizing T cells are CAR-T cells (gene-modified), Tumor-infiltrating Lymphocytes (TILs), and Regulatory T cells (Tregs). Each therapy traditionally uses different reagents to optimize the manufacturing process, as seen below:
T cell-based Cell Therapies

CAR-T Cells

T cells are commonly isolated via leukapheresis, but also can be differentiated from stem cells.

Activation: Anti-CD3 Antibodies and Anti-CD28 Antibodies bound to porous plates or onto artificial antigen presenting cells (AAPCs).
Residue Detection: Cytokine detection kits, including IL-2, IL-7, and IL-15, are available to ensure no residual growth factors are present in the final therapeutic.

Tumor-infiltrating Lymphocytes

TILs are isolated by digesting tumor tissue with collagenase. The isolated cells are then cultured and activated for use.

Culturing: TIL cells are induced using IL-2-infused culture media.
Activation: Anti-CD3/CD28 Antibodies as well as Anti-CD3/CD28 beads for activation.
Residue Detection: Anti-CD3/CD28 antibody detection kits are used to ensure no activation factors are remaining after TIL purification.

Regulatory T Cells

Treg cells are isolated from peripheral blood, and purified for culturing, activation, and detection.

Culturing: Treg cells, derived from peripheral blood, are induced using IL-2 and TGF-β-added growth factors in culture media.
Activation: Anti-CD3/CD28 antibodies for activation.
Residue Detection: Cytokine detection kits are also used to ensure no additional growth factors make it to the final therapeutic.
