Claudin 18.2 Proteins

Claudin18_2
INTRODUCTION
Claudin is a family of proteins first discovered by Furuse in 1998 , which contains at least 27 members. It’s a type of four-pass transmembrane protein that expressed at the tight junction of epithelium and endothelium which is a component of tight junction strands that play critical roles in maintaining cell polarity and signal transductions. Claudin 18.2, the subtype of Claudin 18, is reported only limitedly expressed in differentiated epithelial cells of the gastric mucosa whereas significantly expressed in a variety of malignant tumor tissues such as gastric cancer, pancreatic cancer, cholangiocarcinoma, ovarian cancer and lung cancer etc. Thus it has emerged as an ideal target for immunotherapy including monoclonal antibody, bispecific antibody, ADC and CAR-T therapy.
Restricted by the surface area of the cell membrane and the potential cytotoxicity of overexpressed membrane proteins, the expression level of recombinant four-pass Claudin 18.2 proteins is much lower than that of soluble proteins. Furthermore, it is necessary to screen suitable detergents or lipids  to maintain spatial conformation and protein activity in the process of extraction and purification. So that makes it very difficult to obtain Claudin 18.2 antigen with high purity, native conformation and activity, which seriously hinders the development of drugs and therapies targeting Claudin 18.2.

Strcture of Claudin 18.2 (AlphaFold structure prediction)
Relying on the technology platforms of multi-pass transmembrane proteins, ACROBiosystems has successfully developed full-length Claudin 18.2-VLP, complete epitope Claudin 18.2-Detergent, and detergent-free Claudin 18.2-Nanodisc proteins. These products have been validated through ELISA/BLI/SPR to exhibit high biological activity, meeting the requirements for drug development. Meanwhile, we offer integrated SPR /BLI analytical service to meet the needs of drug development, assist drug clinical declaration.
